Angel Pharmaceuticals Presents Latest Translational Research Findings on CPI-818 (ITK Inhibitor) at the 2023 AACR Annual Meeting
New research demonstrates that CPI-818 can successfully modulate the immunosuppressive tumor microenvironment, significantly inhibit the growth of solid tumors and lymphomas in preclinical models, and shows synergistic effects when combined with marketed immune checkpoint inhibitors
Jiaxing, China and Burlingame, Calif., U.S.A. – April 14, 2023 – Angel Pharmaceuticals, a clinical-stage innovative drug company, today announced a new breakthrough for CPI-818, a global first-in-class ITK inhibitor being co-developed with Corvus Pharmaceuticals. This targeted inhibitor, the world's first of its kind, received clinical trial approval from the NMPA Center for Drug Evaluation in 2021 for the treatment of relapsed/refractory T-cell lymphomas. The latest research shows that CPI-818 can successfully modulate the immunosuppressive tumor microenvironment, significantly inhibit the growth of solid tumors and lymphomas in preclinical models, and demonstrates synergistic effects when combined with marketed immune checkpoint inhibitors. The latest translational research findings on this inhibitor as an innovative molecule for tumor immunotherapy will be presented as a poster jointly by Angel Pharmaceuticals, Corvus, and Peking University Cancer Hospital at the AACR (American Association for Cancer Research) Annual Meeting on April 14.
According to the disclosed findings, the inhibitor can successfully modulate the immunosuppressive tumor microenvironment and significantly inhibit the growth of solid tumors and lymphomas in preclinical models. In experiments on the CT26 colon cancer model, EL4 T-cell lymphoma model, and renal cancer model, CPI-818 at doses of 10–30 mg/kg significantly inhibited tumor growth. In animal model studies, CPI-818 not only demonstrated antitumor activity as a monotherapy but also showed synergistic effects when combined with marketed immune checkpoint protein inhibitors. The triple combination therapy of CPI-818 with suboptimal doses of anti-PD-1 and anti-CTLA-4 antibodies achieved complete tumor clearance in 100% of mice in the CT26 colon cancer model. More importantly, this treatment generated antitumor immune memory and durable immunity, rendering the treated mice resistant to tumor rechallenge. In addition to discovering the prominent efficacy of this inhibitor, the researchers further elucidated the molecular and cellular mechanisms underlying CPI-818's antitumor effects. The inhibitor increases CD8 T-cell infiltration into tumors and increases the CD8-to-regulatory T-cell ratio in vivo, thereby enhancing the antitumor activity of cytotoxic T cells and achieving antitumor effects. Therefore, this inhibitor has the potential to become a leading first-in-class drug with applications in both targeted tumor therapy and tumor immunotherapy.
